Ynysmeudwy were defeated 4-0 after they missed their chance to equalise before the break after the visitors had taken an early lead.
After the break, Ynysgerwyn went further ahead and put the game out of reach to ensure Athletic's unbeaten run came to an abrupt end.
Athletic are away next Saturday to Seven Sisters, kick off 2.30pm.
Cwmamman 3 Ynysmeudwy Reserves 3
The Reserves played away against Cwmamman United, and earned a very well deserved draw with two goals from Daniel Picton and one from Chris Jarvis.
Reserves host Longford in the second round of the Open Cup on Saturday (KO 2pm).
